Negative staining is a specialized method used to observe particulate substances or biomacromolecules in samples under a microscope. Once the sample has been purified, the degree of purification is assessed to determine suitability for further electron microscopy analysis. However, traditional biochemical assays may fall short in this regard. Even in cases where complexes maintain their structural integrity, they might represent a blend of subcomplexes with varying compositions or homologous complexes exhibiting diverse conformations. Therefore, negative staining serves as a fast and effective method for the quality assessment of purified biological samples under the microscope. This screening aims to qualitatively assess particle composition and conformational homogeneity, which can only be performed under a microscope.
During the process, protein particles are adsorbed onto a supporting grid with a hydrophilic membrane layer. Next, the area around the protein particles was stained with heavy metal salts to enhance the contrast of the background. Due to the fact that protein particles themselves do not react with heavy metal salts, they can be clearly visualized under an electron microscope, enabling precise observation of protein structure.
